Pulsed focused ultrasound (pFUS) has been successfully used as a noninvasive treatment of solid malignancies in a clinical setting. pFUS destruction leads to tumor debris being presented to immune cells at the tumor microenvironment (TME) inducing an immune response. To date, the molecular changes in the TME following pFUS are poorly understood. In this study, we performed transcriptomic analysis and explored the temporal expression of cytokines, chemokines and trophic factors (CCTF) in TME of mouse xenograft B16 melanoma and 4T1 breast tumor models following pFUS treatment. Ultrasound-image-guided pFUS (VIFU 2000, Alpinion) was administered at 1MHz and 6MPa peak negative pressures.
