Ras proteins transduce signals between cell surface receptors and intracellular signaling cascades. K-Ras4B is particularly frequently mutated in lung, colorectal, and pancreatic cancers, and importantly, its drug resistance is often accompanied by overexpressed YAP1. RASSF5 is an effector which has a critical interactions with K-Ras4B and with MST1/2 in the Hippo Pathway - inhibit MST1/2 resulting in YAP1 overexpression. We propose to figure out the complex of K-Ras4B-RASSF5 mechanism and to reveal how K-Ras4B regulates the Hippo pathway through RASSF5.
