Abstract

NIH and CBER investigators can purchase >8,900 different mouse ES cell lines carrying specific gene knockouts currently for $1470 for the first clone, and $805 each for additional clones of the same target (http://komp.org/fees.php). >10,500 KO vectors for targeting recombination in ES cells, and >1000 KO lines of mice/sperm are available. ~312 mouse strains have been produced and their phenotypic data is available (www.kompphenotype.org). Data for many more mouse lines will be added this year from the KOMP2 program (mousephenotype.org). Many of the KO genes have annotations in www.ncbi.nlm.nih.gov/omim/ (OMIM) via an Advanced Search (www.komp.org/advancedSearch.php). Many of the KOed genes may be embryonic lethals when homozygous. Several vector constructs (diagrammed at komp.org/alleles.php#deletion-csd) are used, and conditional vectors allow tissue-specific or cell-type-specific perturbations by temporally or spatially restricting expression of the KO vector phenotype via combination with various patterns of Cre-recombinase expression. KOMP2 from 2011-2016 is converting the IKMC knockout ES cell libraries into 2,500 lines of mice that have comprehensive phenotypic data. KOMP2 has co-funding from many NIH ICOs and the Common Fund. www.komp.org/ will be the source to obtain mice or sperm, and mousephenotype.org/ will provide access to the comprehensive phenotypic data and protocols.
