Abstract

The Collaborative Protein Technology Resource (CPTR) aims to bring cutting-edge protein-based technologies to the NCI CCR community to facilitate basic and translational research. The CPTR offers mass spectrometry analyses and capillary immunoassays. Although mass spectrometry is a powerful technique, experimental design and sample preparation are critical for a successful outcome. Our experience in mass spectrometry has been successfully combined with the biological and biochemical expertise of our collaborators to identify proteins in complexes or organelles and perform quantitative proteomic analyses (e.g. SILAC labeling). Determination of the composition of protein complexes and organelles is critical for understanding their biological functions. Additionally, determination of the relative quantitation of protein levels following treatment, after protein knock-down, or in different disease states adds critical functional information. Mass spectrometry is also one of the leading methods for determining sites and types of modifications on proteins. These modifications are central to regulation of signaling pathways and protein function. By making available our expertise in mass spectrometry, we seek to help researchers with their proteomics experiments, beginning with initial experimental design and sample preparation and continuing through data interpretation.
