Examination of T cell function is one of the most frequently used output readings in immunological research laboratories around the world. Here, we review multiple high resolution flow cytometry techniques for the study of T cell activation. These techniques include immunophenotyping, viability analysis and proliferation monitoring. Conventional cytometers often lack the resolving power to differentiate the activated and unactivated T cell populations by light scatter detection and quantitative surface antigen measurement. Using a novel cytometer, which measures light scatter and surface antigen parameters at high resolution, we discriminated proliferating and non-proliferating populations with more accurate differentiation than commonly reported. High resolution in flow cytometric data enables better distinction between T cell activation states.
